Experimental principle

The method for measuring the mass of the cells includes a nitrogen determination method, a DNA method, a measurement of a cell dry weight method, a measurement of a wet cell weight method, and a physiological index measurement method.
In order to learn the growth law of microorganisms and improve the characteristics of microbial growth and rapid reproduction, yeast, actinomycetes and cucumber Fusarium oxysporum were used as experimental materials. The experiment used dry/wet weight method, blood cell counting method for direct counting and turbidity. 1. The blood cell counting plate is a commonly used direct micro-technical method, which has the advantages of being intuitive, quick and easy to operate, but it is laborious, hurts the eyes, and requires high concentration of the suspension, generally not too low or too high (usually greater than 10) ~6 / m), the OD value should be between 0.1-0.65, which is difficult to grasp. Moreover, this method can not measure the number of viable bacteria. If a staining solution such as phenol blue is used, the dead cells can be dyed blue, and the living cells are not stained, thereby measuring the number of viable cells.
2. The wet weight method and the dry weight method can reflect the amount of biomass in the fermentation broth, but it does not reflect the physiological state of the hyphae, the degree of aging, or even the differentiation of cells.
3. It is simple and convenient to measure the growth status of the filamentous fungi grown by radiation, but it seems to be not very applicable.
Therefore, each method has its own advantages and limitations. Only after considering the relationship between these factors and the problems that need to be solved, can we choose the better ones or several methods at the same time.
