AAF fixative (alcohol acetate formalin mixed fixative) instruction manual

AAF fixative instructions ?

Key words: fixative, buffer, biochemical solution, Qiaoyu biological reagent

Product introduction :

The purpose of fixation is to preserve the original morphological structure of cells and tissues. The fixative can prevent endogenous lysosomal enzymes from autolysis of their tissues and cells, and inhibit the growth of bacteria and molds. The immobilizing agent inactivates the enzyme by coagulation, formation of an added compound, and the like, thereby changing the internal structure of the protein. The fixative physically changes the extracellular components of the nucleus. The living cells are wrapped in an impermeable membrane before being fixed, and the fixative can destroy the barrier, allowing the macromolecular substance to penetrate the membrane and escape.

The choice of fixative is extremely important for tissue staining and immunohistochemical staining. Usually, several fixatives are mixed and formulated into a composite fixative, which can adapt to the preservation of various tissues and cellular components. However, there is currently no standard fixative. Suitable for the preservation of all tissues and cellular components. The fixative is mainly divided into an aldehyde fixative, a mercury fixative, an alcohol fixative, an oxidant fixative, a picrate fixative, etc., and is commonly used in formalin and alcohol in aldehydes. Ethanol. ?

AAF fixative, also known as alcohol formalin mixed fixative or FAA fixative, consists mainly of ethanol (A), acetic acid (A), and formaldehyde (F), and has both dehydration effects.

AAF fixatives are commonly used for rapid dehydration and fixation as well as rapid fixation of frozen sections. ?

Operation steps (for reference only):

1. Operate according to the specific requirements of the experiment. ?

2. Take fresh tissue into the AAF fixative and place it in the refrigerator for 10 to 30 minutes at low temperature. ?

3. If the conventional paraffin section is not satisfactory due to poor fixation, it can be fixed with AAF fixative for 5 min before dyeing, and the staining effect will be significantly improved. ?

Precautions:

1. AAF fixative has certain damage to the human body. Please operate carefully in a well ventilated environment to avoid inhalation. ?

2. The pH of the fixative is preferably close to neutral and the pH range is 6-8. ?

3. The thickness of the tissue is different, and the fixing time is also different. The proper material selection for the tissue is conducive to the penetration of the fixing solution. Conventional biopsy tissue is suitable for a thickness of 2 to 4 mm, and generally no more than 6 mm. ?

4. The capacity of the fixative should be sufficient. Generally, the volume ratio of fixative to tissue block should be greater than 10:1. If the volume is not large enough, you can replace the fixative 1 or 3 times during the fixation period. ?

5, the effect of temperature on the fixation is very obvious, increase the temperature can accelerate the fixation, but the temperature should not be too high. ?

6. After removing fresh tissue, it should be fixed in time. If it cannot be fixed in time, it should be stored in physiological saline for timely inspection. ?

7. For your safety and health, please wear a lab coat and wear disposable gloves. ?

Validity: ?6 months effective
